摘 要: | 甘肃马铃薯(2.2kg)经缓冲液提取、硫酸铵分级沉淀、DEAE-纤维素、CM-纤维素离子交换、SephadexG150,Senharose6B葡聚糖凝胶层析纯化、水透析和冻干后制得凝集素,总活力为495120u,比活为6641.89u/mg;经聚丙烯改胺凝胶电泳证明是均一的;氨基酸分析表明富含天冬氨酸、谷氨酸、亮氨酸和赖氨酸。

Extraction and purification of the lectin from potato tubers |

Abstract: | The extraction and purification of potato lectin (2. 2 kg) were described. The acetic saltbuffer was used as extracting solvent. The extract was chromatographied with DEAE-cellulose column,CM-cellulose column, G-150 dextrane gel and 6B dextrane gel. The precipitation was dialysed in water andfreeze-dried. The general activity of the potato lectin is 491520 units and specitic activity is 6641. 89 unitsper milligram. The potato lectin was homogeneous indentified by polyamide gel electrophoresis. Asparticacid is the most abundant aminoacid in potato lectin. |
