红车轴草异黄酮对去卵巢大鼠骨质疏松防治作用的实验研究

摘    要:目的探讨红车轴草异黄酮对去卵巢大鼠引起的大鼠骨质疏松症的防治作用。方法采用切除大鼠双侧卵巢方法建立大鼠骨质疏松模型,以尼尔雌醇作阳性对照,采用放射免疫法检测实验大鼠血清雌二醇(E2)、促黄体生成素(LH)、骨钙素(BGP)、白细胞介素6(IL-6),用生物化学法检测大鼠血浆抗酒石酸盐酸磷酸酶(TRAP)。同时用定量超声检测大鼠股骨近端超声振幅衰减(BUA)及大鼠股骨骨干重和骨灰重。结果红车轴草总异黄酮可显著提高E2水平,降低LH及BGP、TRAP和IL-6活性,显著提高实验大鼠股骨干重、灰重。同时大鼠股骨近端BUA明显升高。结论红车轴草异黄酮具有提高雌激素作用,可有效抑制骨吸收,降低骨转化率,加强成骨细胞活性,增加骨密度。对去卵巢引起的大鼠骨质疏松症具有明显的防治作用。

Abstract:Objective To investigate the prevention and treatment effects of Trifolium Pratens L Isoflavones on osteoporosis of ovariectomized rats.Methods Rat models of osteoporosis were induced by ovariectomy,and treated with Trifolium Pratens L Isoflavones in experimental groups and nilestriol in positive control group.Serum levels of estradiol(E_2),luteinizing hormone(LH),osteocalcine(BGP) and interleulin-6(IL-6) of rat models were measured by radioimmunoassay.Serum levels of tartrate resistant acid phsphatase(TRAP) were tested.The broadband ultrasonic attenuation(BUA) of proximal femurs,fat-free dry weight and ash weight of the femur of the rat model were measured by quantitative ultrasound.Results Trifolium Pratens L Isoflavones significantly increased the serum level of E_2 and BUA of proximal femurs,decreased the serum level of LH and BGP or TRAP and IL-6,and increased fat-free dry weight and ash weight of the femur.Conclusions Trifolium Pratens L Isoflavones can prevent and treat osteoporosis in ovariectomized rats by increasing serum estrogens,inhibiting bone resorption,decreasing bone turnover rate,improving activities of osteoblasts and enhancing bone mineral density.
