摘    要:
目的探讨肿瘤坏死因子α(TNFα)抗体在大鼠急性出血坏死性胰腺炎时对脑组织损害的保护作用,从而为胰性脑病的治疗提供实验依据.方法将60只SD大鼠随机分为正常对照组,急性出血坏死性胰腺炎诱导组和TNFα抗体处理组,经胰胆管逆行注射5%牛磺胆酸建立大鼠ANP模型,测定血的TNFα水平,12 h后处死动物,检查胰腺组织形态学变化,并检测脑组织含水量、脑微血管内白细胞聚集及附壁现象.结果经TNFα抗体处理后,血清中TNFα水平、脑组织含水量、脑微血管内白细胞聚集及附壁现象显著降低(P<0.05),胰腺组织病理损害程度减轻.结论TNFα抗体可以减轻急性出血坏死性胰腺炎时脑损害的发生和发展.

Objective:To study the protective effects of tumor necrosis factor α (TNF α) antibody on pancreatic encephalopathy in rats. Methods: 120 SD rats were randomly divided into the normal control group, acute necrotizing pancreatitis induction group and TNF α antibody treated group. Acute hemorrhagic necrotizing pancreatitis model in rats was induced by retrograde injection of 5% sodium taurocholate into the pancreatobiliary duct. Serum TNF αwas detected and animals were killed 12 h after drug administration. The changes of brain water contents, MDA content, leucocytes accumulation and adhesion of microvessels were measured. Results: In group of TNF α antibody treated, serum TNF α level decreased, brain water content, MDA content and leucocytes accumulation and adhesion decreased significantly compared to those of acute necrotizing pancreatitis induction group ( P < 0.05 ). Conclusions:TNF α antibody can alleviate the brain damage in acute hemorrhagic necrotizing pancreatitis.
