摘    要:目的建立抗病毒颗粒(金银花、连翘、蒲公英等)HPLC指纹图谱,并同时测定新绿原酸、绿原酸、隐绿原酸、咖啡酸、木犀草苷、异绿原酸B、3,5-二咖啡酰奎宁酸、4,5-二咖啡酰奎宁酸、蒙花苷的含有量。方法该药物水提液的分析采用ZORBAX Eclipse XDB-C18色谱柱(4.6 mm×250 mm,5μm);流动相乙腈-水(含0.1%甲酸),梯度洗脱,体积流量1.0 mL/min;检测波长320 nm;柱温30℃。结果 12批样品指纹图谱中有17个共有峰,并指认出其中10个,相似度大于0.97,无明显分类。9种成分在各自范围内线性关系良好(R2>0.999 0),平均加样回收率97.34%~102.69%,RSD 0.79%~2.20%。结论该方法准确、重复性好,可用于抗病毒颗粒的质量控制。

Abstract:AIM To establish the HPLC fingerprints of Kangbingdu Granules(Lonicerae japonicae Flos, Forsythiae Fructus, Taraxaci Herba, etc.) and to simultaneously determine the contents of isochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, caffeic acid, hibiscus, isochlorogenic acid B, 3,5-dicaffeoylquinic acid, 4,5-dicaffeoylquinic acid and buddleoside.METHODS The analysis of aqueous extract of this drug was performed on a 30 ℃ thermostatic Agilent Eclipse XDB-C18 column(4.6 mm×250 mm, 5 μm), with the mobile phase comprising of acetonitrile-water(containing 0.1% formic acid) flowing at 1.0 mL/min in a gradient elution manner, and the detection wavelength was set at 320 nm.RESULTS There were seventeen common peaks in the fingerprints of twelve batches of samples, ten of which were identified with the similarities of more than 0.97 and without clearly classification. Nine constituents showed good linear relationships within their own ranges(R2>0.999 0), whose average recoveries were 97.34%-102.69% with the RSDs of 0.79%-2.20%.CONCLUSION This accurate and reproducible method can be used for the quality control of Kangbingdu Granules.
