摘    要:
目的建立HPLC法同时测定桑白皮Morus alba L.中绿原酸、二氢桑色素、氧化白藜芦醇、桑辛素O、桑根酮C的含有量。方法桑白皮甲醇提取物的分析采用Dimonsiol C18色谱柱(4.6 mm×250 mm,5μm);流动相乙腈-0.2%醋酸,梯度洗脱;体积流量1.0 mL/min;柱温30℃;检测波长340 nm。结果绿原酸、二氢桑色素、氧化白藜芦醇、桑辛素O、桑根酮C分别在3.25~104.00μg/mL(r=0.999 6)、4.81~154.00μg/mL(r=0.999 7)、5.38~172.00μg/mL(r=0.999 5)、1.88~60.00μg/mL(r=0.999 7)、11.19~358.00μg/mL(r=0.999 5)范围内线性关系良好。平均加样回收率分别为98.98%(RSD=1.70%)、100.73%(RSD=1.35%)、99.71%(RSD=1.64%)、100.07%(RSD=1.11%)、99.90%(RSD=2.16%)。结论该方法准确简便,重复性好,可用于桑白皮药材的质量控制。

Abstract:
AIM To establish an HPLC method for the simultaneous content determination of chlorogenic acid,dihydroquinochrome,oxidized resveratrol,moracin O and sanggenon C in Morus alba L..METHODS The analysis of methanol extract of M. alba was performed on a 30 ℃ thermostatic Dimonsiol C18 column(4.6 mm×250 mm, 5 μm), with the mobile phase comprising of acetonitrile-0.2% acetic acid flowing at 1.0 mL/min in a gradient elution manner,and the detection wavelength was set at 340 nm.RESULTS Chlorogenic acid,dihydroquinochrome, oxidized resveratrol, moracin O, sanggenon C showed good linear relationships within the ranges of 3.25-104.00 μg/mL(r=0.999 6), 4.81-154.00 μg/mL(r=0.999 7), 5.38-172.00 μg/mL(r=0.999 5), 1.88-60.00 μg/mL(r=0.999 7),11.19-358.00 μg/mL(r=0.999 5), whose average recoveries were 98.98%(RSD=1.70%),100.73%(RSD=1.35%),99.71%(RSD=1.64%),100.07%(RSD=1.11%),99.90%(RSD=2.16%), respectively.CONCLUSION This accurate,stable and reproducible method can be used for the quality control of M. alba.
