摘    要:
目的:建立一种孕早期、简便且无创伤性的胎儿性别的产前诊断方法。方法:应用降落PCR和二次PCR扩增技术,联合检测妊娠14~20周的正常孕妇血浆中游离胎儿DNA的SRY基因和DXS8377位点。结果:24例孕妇血浆中游离DNA检测SRY基因和DXS8377位点,明确判断性别22例,不能明确判断性别2例。结论:采用降落PCR和二次PCR扩增技术,联合检测孕妇血浆中游离胎儿DNA,可提高孕早期胎儿性别判断准确率。

Abstract:
Objective:To establish an approach to non-invasive prenatal sex identification in early pregnancy.Methods:By dual PCR or touch-down PCR,both SRY gene and X-STR locus were detected from different sources of 24 DNA samples of normal pregnant women whose gestation ages were 14~20 weeks.Results:Fetal sex was identified by both SRY gene and X-STR locus.22 of the 24 cases were identifed clearly,and 2 cases were not identified.Conclusion:Both touch-down PCR and duel PCR asssays are used in detecting of free fetal DNA in maternal plasma at SRYgene and DXS8377 locus.This method can improve the accuracy of fetal sex identification in early pregnancy.
