慢性阻塞性肺病患者IL-4、IL-13以及ADRβ_2基因多态性的检测

摘    要:目的:探讨白介素-4(IL-4)、白介素-13(IL-13)以及β2肾上腺素能受体(ADRβ2)基因多态性与我国汉族慢性阻塞性肺病(COPD)发生之间的相关性。方法:本研究采用病例对照性研究,通过聚合酶链反应和限制性片段长度多态性(PCR-RFLP)方法检测106例COPD患者和72例健康对照者IL-4[-589C/T、-33C/T、可变数目串联重复序列(VNTR)]、IL-13(-1111C/T、-2044G/A)以及ADRβ2(-46A/G及-79C/G)的基因多态性,同时以Taq-Man等位基因识别技术检测单倍型频率、基因型以及等位基因频率。结果:COPD组与对照组的ADRβ2-79C/G基因型的频率分布存在显著性差异(P=0.002)。同时,2组间单倍型基因频率(IL4-589C/T:ADRβ2-79C/G以及IL4VNTR:ADRβ2-79C/G)也存在显著性差异(P值分别为0.033与0.001)。结论:ADRβ2-79C/G基因多态性及单倍型可能参与COPD的发病机制。

Abstract:Objective: To study the relationship between the polymorphism of IL-4,IL-13,or beta 2-adrenoceptor(ADRβ2) and COPD in Chinese Han population.Methods: A case-control association study was conducted on 106 patients with COPD and 72 control subjects.The following polymorphisms were genotyped by polymerase chain reaction-restricted fragment length polymorphism(PCR-RFLP):-589 C/T,-33 C/T,and variable number of tandem repeat(VNTR) in IL-4,-1111C/T and-2044 G/A in IL-13,and-46 A/G and-79 C/G in ADRβ2.Pairwise haplotype frequencies as well as genotype and allele frequencies were analyzed by using TaqMan allelic discrimination technique.Results: The distribution of the genotype frequencies of ADRβ2-79 C/G was significantly different between COPD and control groups(P=0.002).Also,the distributions of the haplotypes(IL-4-589 C/T:ADRβ 2-79C/G;IL-4 VNTR: ADRβ2-79 C/G)(P=0.033 and P=0.001 respectively) showed significant differences between COPD and control groups.Conclusion: The ADRβ2-79 C/G polymorphism and the haplotypes shown in this study may be involved in the pathogenesis of COPD.
