甲基莲心碱对氧化低密度脂蛋白导致平滑肌细胞脂质堆积和增殖的影响

摘    要:目的 观察甲基莲心碱(Nef)对氧化低密度脂蛋白(oxLDL)导致兔主动脉平滑肌细胞(VSMC)内脂质堆积、增殖的影响。方法 以oxLDL为泡沫细胞诱导剂,测定细胞内超氧化物歧化酶(SOD)及谷胱甘肽过氧化物酶(GSH—PX)的活性。观察Nef对oxLDL导致的VSMC增殖及测定细胞内游离Ca^2+浓度。结果 Nef可显著抑制oxLDL诱导的VSMC内甘油三酯、总胆固醇堆积,oxLDL组细胞内SOD和GSH—PX活性较正常组降低,Nef处理组VSMC内SOD和GSH—PX活性明显升高(P〈0.05)。Nef可显著降低VSMC内Ca^2+。结论 Nef可显著抑制oxLDL诱导VSMC的脂质堆积和增殖。

Abstract:Objective To investigate the effects and mechanisms of neferine on lipids accumulation and proliferation of vascular smooth muscle cells (VSMC) induced by oxLDL. Methods VSMC were exposed to oxLDL, and then incubated with different concentrations of neferine respectively. The contents of tryglyceride (TG) and total cholesterol (TC) were measured and the activities of SOD and GSH-PX were detected in VSMC. The proliferation of VSMC and concentration of intracellular calcium were assayed by MTT and Fura-2 fluorescence method. Results Neferine could inhibit the intracellular lipid accumulation in VSMCs induced by oxLDL. At the same time, the activities of SOD and GSH-PX in VSMC were increased. It was also found that neferine inhibited VSMC proliferation induced by oxLDL. Neferine treatment could significantly decrease the content of intracellular calcium. Conclusion Neferine can inhibit foam formation and proliferation of VSMC. The mechanisms of these phenomena may be associated with the increase of self-anti-oxidation ability and the inhibition of intracellular calcium of VSMC.
