PMS肝气逆证模型大鼠中枢5-HT_(1A)R结合活性分析及其药物干预

目的:检测PMS肝气逆证模型大鼠大脑皮层和海马5-HT1AR(5-羟色胺1A受体)结合活性的变化,探讨中药经前平颗粒治疗PMS肝气逆证的作用机制。方法:电刺激复制PMS肝气逆证大鼠模型。放射性配体受体结合实验测定大鼠皮层和海马5-HT1AR结合活性变化。结果:与正常组比较,肝气逆证造模组大鼠大脑皮层5-HT1AR结合活性显著上调(P<0.01),海马5-HT1AR结合活性上调但不显著(P>0.05),经前平颗粒可显著下调肝气逆证模型大鼠上述脑组织内5-HT1AR结合活性。结论:PMS肝气逆证模型大鼠的大脑皮层和海马5-HT1AR结合活性上调。中药经前平颗粒可能通过调节5-HT1AR结合活性发挥治疗PMS肝气逆证的作用。

Analysis on the binding activity of 5-HTl1R in the neural of PMS model rats with liver-Qi invasion and the medicine effects

Objective:To explore the mechanism of Jingqianping granule acting on the liver-Qi invasions by detecting changes of the binding activity of 5-HT1AR in the pallium and hippocampus of PMS model rats with liver-Qi invasion.Methods:To prepare PMS model rats with liver-Qi invasion by electric stimulating,and to detecte the specific binding of 5-HT1AR in the membrane preparation of several brain regions by radioligand combination experiment with receptor.Results:Compared with the normal group,the specific binding activity of 5-HT1AR in the pallium of liver-Qi invasion model rat upregulated markedly(P<0.01),and 5-HT1AR in hippocampus increased but insignificantly(P>0.05).Compared with the model group,the binding activity of pallium 5-HT1AR in Jingqianping granule group downregulated,hippocampus 5-HT1AR downregulated markedly.Conclusions: The binding activity of 5-HT1AR in the pallium and hippocampus of PMS model rats with liver-Qi invasion upregulated compared with the normal group.And Jingqianping granule may treat PMS with liver-Qi invasion by regulating the binding activity of 5-HT1AR.
