湖南地区玉竹根腐病病原鉴定及其生物学特性研究

摘    要:目的:明确导致湖南地区玉竹根腐病发生的主要病原。方法:通过调查湖南玉竹主产区玉竹根腐病发生情况,对病原进行分离,并采用柯赫氏法则进行致病性测定,最后根据真菌形态和分子鉴定结果确定病原菌种属,同时对该病原菌基本的生物学特性进行了测定。结果:芬芳镰刀菌Fusarium redolens为玉竹根腐病主要病原,该菌菌丝生长适宜碳源为蔗糖,氮源为牛肉膏,温度为25℃左右,pH为8~9。结论:本研究明确了湖南玉竹主产区玉竹根腐病病原及其基本的生物学特性,不但为该病害的防控提供了依据,还可为抗病育种奠定基础。

Abstract:Objective: To confirm the pathogen of Polygonatum odoratum root rot in Hunan, China. Methods: In this study, Fungi was isolated by common method, Pathogenicity was tested according to koch' s postulates, and identification of the fungi were carried out by the method of morphology and molecular biology. The effect of temperature, pH, carbon and nitrogen sources on colony growth was also assessed by the growth rate. Results: Fusarium redolens was identified as the pathogen of Polygonatum odoratum. The results showed that the optimum temperature was about 25 ℃, and the optimum pH for hyphae growth was 8-9. The growth of colony was the best with sucrose as the carbon source, and beef extract as the nitrogen source. Conclusion: This study confirmed the pathogen of Polygonatum odoratum root rot in Hunan and its biological characterization, which can provide evidence for prevention and treatment of this disease, also benefit to the breeding and disease resistance of Polygonatum odoratum.
