摘    要:目的:探讨细胞膜色谱法应用于吉非替尼在肺癌细胞细胞膜上保留性。方法:应用人肺腺癌细胞A549制备肺癌细胞膜固定相,低压湿法装入色谱柱(10mm×2mm,ID)中,在37℃条件下,以5mmol/L磷酸钾缓冲液(pH7.4)为流动相,流速为0.2ml/min,在253nm检测波长下应用高效液相色谱检测吉非替尼在肺癌细胞膜的保留特性。结果:吉非替尼在肺癌细胞膜上有保留。结论:吉非替尼在肺癌细胞膜色谱柱中有保留,可进一步进行研究。

The initial investigation of gefitinib chromatographic retention on lung cancer cell membrane

Abstract:Objective: To investigate Gefitinib chromatographic retention on lung cancer cell by cell membrane chromatography. Methods:A cell membrane stationary phase (CMSP) was prepared by immobilizing human pulmonary adenocarcinoma cell hne A549 on the surface of a silica carrier In this model, the column ( 10 mm×2 mm, I. D. ) packed with CMSP, 5 mmol/L potassium phosphate buffer solutions ( pH 7.4) as mobile phase with the flow rate of 0.2 mL/min at 37.5 ℃were used in the following studies. The retention characteristic of the Gefitinib was investigated under the chromatographic conditions above at the wave length of 253nm. Results: Gefitinib can retain on the lung cancer cell membrane. Conclusion: Gefitinib can retain on the lung cancer cell membrane chromatographic column, and can be further take into research.
