瑶药葫芦钻提取物抗肿瘤作用的实验研究

摘    要:目的研究葫芦钻提取物的抗肿瘤作用。方法体外抗肿瘤活性研究采用MTT比色法,靶细胞采用人肝癌细胞BLE-7404;通过复制小鼠S180,H22腹水瘤和肉瘤动物模型,观察葫芦钻提取物对小鼠平均生命延长率和肿瘤抑制率的影响来评价体内抗肿瘤作用。结果葫芦钻水、醇提取物浓度为20 mg/ml时,对体外培养的肿瘤细胞的抑制率均>50%;葫芦钻水、醇提取物的最大耐受剂量均是150 g/kg;经3次实验,葫芦钻水、醇提取物高、低剂量组对S180腹水瘤小鼠平均生命延长率均>50%,葫芦钻醇提物高剂量组对H22腹水瘤小鼠平均生命延长率>50%。葫芦钻水、醇提取物高、低剂量组对S180肉瘤小鼠平均瘤重抑制率均>30%,对H22肉瘤小鼠平均瘤重抑制率均>50%。结论葫芦钻水、醇提取物具有一定的抗肿瘤作用。

Abstract:Objective To investigate the antitumor activity of Yao Herb - - Huluzuan. Methods The antitumor activity in vitro against human cancer cell lines - - hepatocarcinoma BEL -7404 was determined by MTT method. The antitumor activity in vivo was evaluated by the prolongation ratios of life - span and the inhibition ratios of the tumor weight of transplanted mouse S180, H22 solid and ascites carcinoma model fed on extracted substances from Huluzuan. Results When the dose of water, alcohol extracted substances from Huluzuan was 20mg/ml, the inhibition ratios of the tumor cell in vitro was in excess of 50%. Both maximum tolerated dose of water, alcohol extracted substances from Huluzuan were 150 g/kg. In three times of the experiment, prolongation ratios of life-span at high and low dose of water, alcohol extracted substances exceed 50% on mice S180 ascites carcinoma model in average, and at high dose of alcohol extracted substances exceeded 50% on mice H22 ascites carcinoma model in average, inhibition ratios at high and low dose of water,alcohol extracted substances exceed 30% on mice S180 solid carcinoma model and exceed 50% on mice tt22 solid carcinoma model in average. Conclusion Water, alcohol extracted substances from Huluzuan has antitumor activity to some extent.
