脑胶质瘤/树突状细胞融合瘤苗的制备与鉴定

摘    要:
目的:制备树突状细胞(DC)融合瘤苗,观察其防治胶质瘤的疗效,并探讨其作用机制。方法:诱导正常人外周血来源DC,在PEG作用下将其与原代培养胶质瘤细胞融合,通过双荧光标记法检测融合率、MTT比色法检测融合疫苗体外杀伤脑胶质瘤细胞的细胞毒作用。结果:融合瘤苗免疫荧光检查阳性,其激活的CTLs对胶质瘤的杀伤作用显著高于对照组,而且杀伤活性随着效靶比的增加而增加。结论:PEG法可有效制备DC与胶质瘤细胞融合瘤苗,融合瘤苗是一种更为有效的以DC为基础的抗胶质瘤方法。

Abstract:
Objective:To prepare the fusion cells of glioma cell and dendritic cells and study the anti-tumor effect and mechanism of fusion vaccine of DCs and glioma cells.Methods: DC derived from normal peripheral blood are induced and fused with glioma cells cultured primarily under PEG.The fusion ratio is determined by two-colour fluorescent labeling,Cytotoxicity is determined by MTT method.Results: Immunolluorescence showed positive in fusion cells.The CTL activity against glioma cells induced by DC/glioma fusion cells was significantly higher than that induced by coculture of DC and glioma fusion cells as well as other control groups.The Cytotoxicity was increased as the ratio of effectors and target cells increased.Conclusion: PEG chemical fusion method is an effective way to prepare the fusion of glioma cell and dendritic cells.The fusion cell is a more effective anti glioma strategy among those based on DC.
