丹酚酸A对缺氧/复氧损伤条件下脑微血管内皮细胞ICAM-1表达的影响

摘    要:[目的]通过对观察丹酚酸A对缺氧/复氧(H/R)损伤条件下脑微血管内皮细胞细胞间黏附因子-1(CAM-1)表达的影响,探讨丹酚酸A对缺血再灌注急性期脑保护作用机制.[方法]培养BMEC,丹酚酸A预处理20 h后制作(H/R)/模型,逆转录-聚合酶链式反应(RT-PCR)法检测BMEC细胞间黏附因子-1 ICAM-1 mRNA的表达.以尼莫地平作为阳性对照药.[结果]模型组与空白对照组相比ICAM-1 mRNA表达明显升高(P<0.01).丹酚酸A组ICAM-1 mRNA表达低于模型组(P<0.05).[结论]丹酚酸A可降低H/R损伤条件下BMEC ICAM-1 mRNA表达的影响,提示阻抑粒细胞黏附作用是其脑保护作用机制之一.

Abstract:[Objective] To investigate the action of Sal A on the expression of cellular adhesion molecules after hypoxia/reoxygenation (H/R) injury in brain microvascular endothelial cells (BMEC). [Methods] After pretreating with Sal A for 20h, the H/R was successfully established H/R on BMEC. Then the expression of ICAM-mRNA was evaluated by RT-PCR method.
