摘    要:
 目的建立快速、灵敏的高效液相色谱-荧光检测法测定人血清中拉呋替丁药物浓度。方法以SupelcosilTMLC-18-DB(4.6mm×250mm,5μm)为色谱柱,[甲醇-(醋酸-醋酸钠缓冲液)](pH4.5)=1∶2为流动相,荧光检测波长:EX=285nm,EM=313nm。结果测定血清中拉呋替丁方法的线性范围为2.5~300μg·L-1,r=0.9997(n=5),定量下限为2.5μg·L-1。日内、日间精密度(RSD)均小于12%。结论本方法操作简便、灵敏度高,可满足拉呋替丁药动学研究需要。

Abstract:
OBJECTIVE To develop a quick and sensitive HPLC asssy for the determination of lafutidine in human serum.METHODS The sample was separated on a SupelcosilTM LC-18-DB(4.6 mm×250 mm,5 μm)column,with a mobile phase of methanol-sodium acetate buffer(1∶2,pH 4.5),and lafutidine was detected by a fluorescence detector with an excitation wavelength of 285 nm and an emission wavelength of 313 nm.RESULTS The linear range of lafutidine was 2.5~300 μg·L-1(r=0.999 7,n=5).The coefficient of variation within day and between days did not exceed 12%.CONCLUSION The method appeared to be simple,convenient and precise for the determination of lafutidine concentration and clinical pharmacokinetic study of lafutidine.
