胰胆管连接异常动物模型的胰型淀粉酶同工酶分析

摘    要:目的 观察胰胆管连接异常(PBM)模型血清和胆汁胰型淀粉酶同工酶的变化。方法 采用十二指肠—胆囊吻合术建立猪PBM模型(n=4),收集手术前、手术后7d、14d、30d、60d血清及胆汁;采用胰管结扎法建立猪急性胰腺炎模型(n=2),取血清。应用不连续缓冲系统薄膜电泳法行酶谱分析。结果 IBM模型在实验第60d时的总胆管无明显扩张。术前血清中胰型淀粉酶同工酶仅出现P1、P2波峰;7d后出现异常的P3、P4、P5、P6波峰,无优势波峰。术后14d、30d、60d的血清胰型淀粉酶同工酶出现率与术后7d时相似,而P6略多见;术后血清胰型淀粉酶同工酶出现率和构成比与胆汁相似。急性胰腺炎组仅出现P1—P3波峰。结论 PBM动物模型的血清和胆汁中均存在异常的胰型淀粉酶同工酶,该异常与急性胰腺炎不同,具有特异性。

Abstract:Objective To investigate the changes of pancreatic amylase isoenzymes in serum and bile from a pig model with pancreaticobiliary maljunction(PBM).Methods On four young pigs,pancreatico-duodeno-cholecystostomy was performed to establish the PBM model.Serum was taken before operation and at days7,14,30and60postoperatively.The pancreatic ducts of pigs were ligated to create an acute pancreatitis model.Electrophoresis in discontinuous buffer system was applied to fractionalize the pancreatic amylase isoenzymes.Results PBM model had no obviously choledochectasia at60-day There were peaks of P1and P2band only in the serum pancreatic amylase isoenzymes(PAI)before operation.The peaks of abnormal P3,P4,P5,P6bands were seen at7days after the operation,but predominat peak had no.The identitied PAI rate was similar among at7-day,and at14-day,30-day60-day after operation,but band P6staining was a slightly strong,occurring rate and constitutional ratio in the sera were siminal to those of bile.Peaks of P1-P3bands were identified only in the pancreatitis group.Conclusions There is abnormal pancreatic amylase isoenzyme in serum and bile of PMB animal model,the PAI abnormality bands isdifferent from acute pancredtitis, it has self-specificity.
