摘    要: 目的观察N-甲基-D-天门冬氨酸(N-methy-D-aspartate,NMDA)对体外培养成年大鼠海马神经前体细胞增殖及胞浆内游离钙离子的影响.方法体外传代培养的成年大鼠海马神经前体细胞经碱性成纤维细胞生长因子(bFGF)、NMDA处理后,采用VIT法测定细胞增殖,激光共聚焦显微镜测定细胞胞浆内游离钙离子浓度.结果与对照组相比,NMDA可明显促进成年大鼠海马神经前体细胞增殖和细胞胞浆内游离钙离子浓度的升高(P<0.01),其作用效果呈剂量和时间的效应关系.结论NMDA可促进体外培养成年大鼠海马神经前体细胞的增殖,其可能机制与NMDA受体操控的细胞内Ca2+浓度升高所致的一系列级生物学级联事件的发生有关.

Abstract:Objective To investigate the effect of N-methy-D-aspartate (NMDA) receptors on the cell proliferation and intracellular free calcium concentration of hippocampus neural precursor cells in cultured adult rat in vitro . Methods The passage cultured adult hippocampus neural precursor cells from adult rat in vitro were pretreated by basic fibroblast growth factor (bFGF) and NMDA. Cell proliferation was assessed by MTT colorimetric assay, and intracellular free calcium concentration was examined by confocal microscopy.Results Compared with control group, NMDA significantly increased the proliferation and intracellular free calcium concentration in the cultured adult hippocampus neural precursor cells activated by bFGF in vitro ( P < 0.01), in a concentration -and time - dependent manner. Conclusion The activation of NMDA receptors-Ca2 pathway can promote the proliferation of adult hippocampus neural precursor cells cultured in vitro , and the accom-modational mechanisms of enhanced cell proliferation are possibly related to a series of neurobiological events after the increase in intracellular free calcium concentration.
