载脂蛋白E基因多态性与2型糖尿病的关系

目的:探讨载脂蛋白E基因型与2型糖尿病的易感性。方法:应用multi-ARMS快速分型法对316例T2DM患者、512例健康对照人群的ApoE基因第4外显子112位Cys/Arg和158位Arg/Cys进行检测;随机抽取分型标本进行DNA测序法验证。结果:ε2/2、ε2/3、ε3/3、ε4/2、ε4/3、ε4/4基因型在二组中的频率分布为:0.6%,5.7%,72.8%,1.9%,14.9%,4.1%(T2DM组);0.6%,9.4%,70.1%,1.8%,17.0%,1.2%(对照组)。二组间差异有显著性统计学意义(χ2=11.45,P<0.05),T2DM组ε4/4基因型频率明显高于对照组(4.11%VS1.17%)。结论:ApoEε4/4基因型可能与T2DM的易感性有关。

The Association of Apolipoprotein E Genotype with Type 2 Diabetes Mellitus

Objective:To study whether apolipoprotein E(ApoE)genotype was associated with type 2 diabetes mellitus(T2DM).Method:Multi-ARMS and PCR-SSCP were used to determine the ApoE genotype of 828 Chinese individuals(316 cases of T2DM and 512 healthy controls).The genotyping results were confirmed by DNA sequencing.Results:The distribution of ApoE ε2/2,ε2/3,ε3/3,ε2/4,ε3/4 and ε4/4 were 0.6%,5.7%,72.8%,1.9%,14.9%,4.1% in T2DM group,and 0.5%,9.4%,70.1%,1.8%,17.0%,1.2% in the control group respectively.The ApoE genotype frequencies between the two groups showed significant difference(χ2=11.45,P<0.05).The distribution of ε4/4 in T2DM group was higher than in control group(4.11% VS 1.17%).Conclusion:The results suggested that subjects with ApoE ε4/4 genotype might be susceptible for T2DM.
