肺癌组织中bcl-2和Bax的表达及其与细胞凋亡和增殖的关系

摘    要:目的 :探讨bcl 2基因和Bax基因在肺癌中表达及其与细胞凋亡和增殖的关系。方法 :对 38例肺癌组织及 2 1例肺部良性组织 ,应用原位杂交技术检测了bcl 2mRNA表达 ;用TUNEL技术对细胞凋亡进行了检测 ,用免疫组织化学法对Bax蛋白和PCNA进行检测。结果 :肺癌组织bcl 2基因、Bax蛋白及PCNA的表达明显高于肺部良性组织 ,P <0 .0 1;肺癌组织中凋亡指数和增殖指数均高于肺部良性组织 ,P <0 .0 1;肺癌组织中bcl 2阳性凋亡指数明显低于bcl 2阴性 ,P <0 .0 1;而Bax阳性凋亡指数明显高于Bax阴性 ,P <0 .0 1;它们之间的增殖指数均无差异。肺癌组织中bcl 2 (+) Bax(- )的凋亡指数明显低于bcl 2 (- ) Bax(+) ,P <0 .0 1;它们之间的增殖指数无差异。bcl 2(- ) Bax(- )肺癌组织中的凋亡指数明显高于肺部良性组织 ,P <0 .0 1。结论 :bcl 2、Bax蛋白在肺癌中的表达率较高 ,同时在肺癌中细胞凋亡和细胞增殖亦较活跃 ;bcl 2基因过度表达抑制细胞凋亡 ,Bax蛋白过度表达促进细胞凋亡 ,而它们表达与细胞增殖无明显关系。bcl 2基因、Bax蛋白、细胞凋亡和PCNA很可能在肺癌的发生、发展、诊断、预后和治疗方面起着重要的作用

Abstract:Objective: To explore the expression of bcl 2 and Bax in the lung cancer and the expression of the relationship between bcl 2 and/or Bax to apoptosis and/or proliferation. Methods:In 38 cancer cases and 21 benign respiratory cases, In situ hybrization technique was applied to detect bcl 2 mRNA; TUNEL technique was applied to detect the apoptosis and the immunohistochemistry was applied to detect Bax oncoprotein and PCNA.Results:The amount of bcl 2 mRNA and Bax oncoprotein of lung cancer tissues was increased obviously, P<0.01. The AI and MI of lung cancer were higher than those of the lung benign cases, P<0.01; The AI of cancer tissues with bcl 2 positive was obviously lower than that with bcl 2negative,P<0.01; The AI of cancer cases with Bax positive was obviously higher than that with Baxnegative, P< 0.01 . Their MIs were not distinctly different. The AI with bcl 2(+)/Bax(-) cancer biopsy specimens was lower than that with bcl 2(-)/Bax(+), P<0.01; The AI and MI of bcl 2(-)/Bax(-) lung cancer biopsy specimens were higher than those of benign biopsy specimens. Conclusions: The amount of bcl 2 mRNA and Bax oncoprotein is increased obviously and the apoptosis and proliferation are active in the lung cancer. Bcl 2 and Bax, especially the ratio of bcl 2 and Bax play an important role in the apoptosis in the lung cancer. The expression of bcl 2 can prevent apoptosis while the expression of Bax induces apoptosis. bcl 2,Bax,apoptosis and PCNA can be used in the lung cancer molecular markers.
