摘    要:目的探讨缺血修饰白蛋白(IMA)可否成为多柔比星(DOX)心肌毒性反应的早期诊断指标。方法取乳鼠心肌细胞与6种不同浓度的DOX相互作用,观察DOX导致心肌毒性模型中活性细胞百分率、细胞凋亡百分率情况,并检测细胞培养液中肌酸激酶同工酶(CK-MB)、肌钙蛋白(c Tn T)及IMA变化情况。结果随DOX浓度增加及培养时间延长,活性细胞百分率逐渐下降(F=23.3,P<0.05),细胞凋亡百分率也逐渐下降(F=30.4,P<0.05)。运用SPSS分析软件的斯伯曼/斯皮尔曼相关系数进行分析,结果显示,IMA与细胞凋亡情况在3 h就开始存在相关性(r=0.891,P<0.05),而c Tn T与细胞凋亡情况12 h开始相关(r=0.658,P<0.05);CK-MB与细胞凋亡情况48 h开始相关(r=0.711,P<0.05)。结论 IMA成为DOX心肌毒性反应的早期诊断指标具有实验室基础。

Diagnosis index of doxorubicin myocardial toxicity:a study of ischemia modified albumin
Abstract:Objective To investigate the feasibility of ischemia modified albumin( IMA) to be the early diagnosis index of doxorubicin ( DOX) myocardial toxicity. Methods Neonatal rat cardiomyocytes were interacted with DOX in 6 kinds of different concentrations, the status of cell activity rate and cell apoptosis were observed on DOX induced myocardial apoptosis toxicity models,and the changes of creatine kinase isoenzyme-MB( CK-MB) ,cardiac troponin T( cTnT) and IMA in cell cultures were inspected. Results Along with the increase of DOX concentration and extension of incubation time,the cell activity rate gradually decreased(F=23. 3,P<0. 05), and apoptosis percentage gradually decreased(F=30. 4,P<0. 05). Using berman/szpilman correlation coefficient in SPSS analysis software,the results showed that there was correlation between IMA and cell apoptosis in 3 hours(r=0. 891,P<0. 05). There was cor-relation between cTnT and cell apoptosis in 12 hours(r=0. 658,P<0. 05). There was correlation between CK-MB and cell apoptosis in 48 hours(r=0. 711,P<0. 05). Conclusion There is a laboratory basis for IMA to be the early diagnosis index of DOX myocardi-al toxicity.
