脑梗死患者Sema4D表达与血浆Gas6的水平及意义简

目的 探讨脑梗死急性期患者外周血淋巴细胞Sema 4D mRNA的表达和血浆可溶性Sema 4D（sSema 4D）的水平及临床意义;并探讨血浆sSema 4D水平与Gas 6水平的相关性及其临床意义.方法 检测299例脑梗死急性期患者及195例正常对照者的Sema 4D及Gas 6水平.采用Realtime PCR检测两组对象周围血淋巴细胞的Sema 4D mRNA表达,同时应用酶联免疫吸附法（ELISA）测定其血浆中sSema 4D及Gas 6水平.结果 脑梗死组Sema 4D mRNA 表达值为2.23（1.10,3.76）×10^4IU/ml较正常对照组0.49（0.252,0.78）×10^4IU/ml明显上调,差异有显著意义（P＜0.01）;血浆sSema 4D浓度及Gas 6浓度明显高于对照组（P＜0.01）.血浆sSema 4D和Gas 6无相关性,相关系数为0.072,P =0.308.结论 脑梗死急性期患者外周血淋巴细胞Sema 4D mRNA表达及血浆sSema 4D水平均上调,提示Sema 4D可以反映脑梗死发生.血浆Gas 6水平较对照组明显升高,提示Gas 6可能是血栓性疾病的一个危险因素.血浆Sema 4D与Gas 6两者在脑梗死过程中可能独立发挥作用.

The Expression Level and Significance of Sema 4D and Plasma Gas 6 in Patients with Cerebral Infarction

Objective To explore the expression and clinical significance of Semaphorin4D（ Sema 4D） mRNA in peripheral blood lymphocyte and plasma soluble Sema 4D （sSema 4D）in patients with cerebral in- farction in acute stage; And to investigate the relationship and clinical significance between the plasma levels of sSema 4D and Gas 6 （ growth arrest- specific gene 6） levels. Methods Taking 299 cases of patients with cerebral infarction as the case group while 195 cases of healthy adults were selected as the control group. The mRNA expression of Sema 4D was detected by Real-time PCR, and the sSema 4D and Gas 6 levels in plasma was detected by ELISA. Results The expression of Sema 4D mRNA was increased in patients with acute pe- riod cerebral infarction median 2.23 ×10^4 u/ml than the normal control group median 0.49 ×10^4 u/ml. Two groups showed significant differenee（P 〈0.01 ）. There was significant difference in Sema 4D on platelet sur- face between patients with acute period cerebral infarction and the normal control group. The former was obvi- ously decreased（P 〈0.01 ）. The concentration of plasma sSema 4D and Gas 6 was significantly higher than the control group（P 〈0.01 ）. No correlation between plasma sSema 4D and Gas 6,the correlation coefficient was 0. 072 ,P = 0. 308. Conclusion The Sema 4D expression and plasma leels in patients with acute period cerebral infarction were rise, and the platelet expression were reduce. These may be related to acute cerebral infarction, can tip the occurrence of cerebral infarction. Both plasma Sema 4D and Gas 6 may play a role in the process of cerebral infarction independently.
