高效液相色谱法测定两性霉素B在家兔体内的血药浓度及药动学

摘    要:
目的:建立血浆中两性霉素B浓度的HPLC测定方法,并用此方法研究两性霉素B在家兔体内的药动力学,方法:血浆经处理后,采用PlatinumC18柱以乙腈-0.005mol.L^-1EDTA-2Na(60:40)为流动相,在紫外405nm处检测。结果:两性霉素B浓度在0.05-12.8ug.ml^-1范围内与色谱峰面积呈良好的线性关系,血浆中药物最低检测浓度为0.005ug.ml^-1,高中低三种浓度的日内及日间RSD%(n=5)分别为(0.34-1.71%)和(0.25-6.78)%,方法回收率和绝对回收率分别为(100.03-105.19)%和(92.18-95.21),药动研究表明,家兔静脉注射单剂量的两性霉素B后其体内血药浓度时间数据符合权重为1/C的三室模型。结论:本方法操作简便,结果准确可靠,很好的满足两性霉素B血药浓度测定及药动学的研究。

Abstract:
OBJECTIVE:To establish an HPLC method for the determination of amphotericin B (AmB) in rabbits plasma and investigate the pharmacokinetic characteristics of AmB by intravenous administration. METHODS:Platinum C18 column and acetonitrile-EDTA-2Na(60∶40) as mobile phase, with UV detection at 405 nm.RESULTS:A good linear relationship was obtained between the peak area and the concentration of AmB from 0.05~12.8 μg*ml-1, and the limit of detection was 0.005 μg*ml-1. The RSD of intra-day and inter-day variations were 0.34%~1.71% and 0.25%~6.78% respectively. The absolute and relative recoveries were 92.18%~95.21% and 100.03%~105.19% respectively. The concentration-time data of AmB after intravenous administration were fitted by three compartments with weight of 1/C. CONCLUSIONS:A simple, highly sensitive and selective method for the determination of AmB level in plasma was established,which could be used for its pharmacokinetic studies.
