摘    要:
[目的]研究全反式维甲酸(ATRA)对神经母细胞瘤细胞增殖的影响.[方法]神经母细胞瘤细胞SY5Y经全反式维甲酸(ATRA)处理不同时间后(2、4、6d)分别于显微镜下观察细胞形态学改变及使用MTS方法观察细胞增殖情况.对经ATRA处理4d的细胞进行溴脱氧尿苷(Brdu)染色观察DNA合成及利用流式细胞仪进行细胞周期分析.[结果]与对照组比较,SY5Y细胞经ATRA处理4d或6d后于显微镜下可见明显的轴突延伸,MTS方法显示细胞增殖明显降低.ATRA处理4d时Brdu染色显示DNA合成减少,利用流式细胞仪对细胞周期的分析表明细胞被阻滞在G1期.[结论]ATRA可抑制神经母细胞瘤细胞增殖.

Abstract:
【Objective】To investigate the effect of all-trans retinoic acid(ATRA) on neuroblastoma cell proliferation.【Methods】 Neuroblastoma cells-SY5Y cells were treated with ATRA for different time(2,4,6 days),then morphology of the cells was observed under microscope and cell growth was evaluated with MTS assay.Bromodeoxyuridine(Brdu)staining and cell cycle analysis with flow cytometry was performed in the cells treated with ATRA for 4 days.【Results】 Compared with the control cells,cells treated with ATRA for 4 days or 6 days showed obvious neurite extention under microscope and growth inhibition detected with MTS assay.Further study of cells treated with ATRA for 4 days indicated an decrease of DNA synthesis by Brdu staining and Cell cycle arrest at phase G1 by cell cycle analysis with flow cytometry.【Conclusion】 ATRA treatment inhibited the proliferation of neuroblastoma cells.
