癌痛消颗粒对小鼠急性毒性实验研究

以癌痛消颗粒的水溶剂对小鼠灌胃,观察小鼠死亡情况及相关指标的变化情况来评价其安全性。半数致死量实验以不同浓度的药液及饱和药液对小鼠灌胃,观察2周未见小鼠死亡,无法测出半数致死量。进行最大耐受量实验,以饱和溶液1天内对小鼠灌胃3次。灌胃后小鼠心、肝、脾、肺、肾、睾丸/卵巢等与体重的比值较对照组差异无显著性意义(均P0.05);血常规及肝肾功能指标值差异亦无显著性意义(均P0.05)。灌入药物总量相当于7.95g生药,折合成人临床日用量为164.6倍,符合临床应用及推广要求,对长期毒性实验剂量的设计和药物Ⅰ期临床实验起始剂量的选择具有重要参考价值。
