摘    要:目的:建立不同产地黄芪指纹图谱的分析方法。方法:采用HPLC-ELSD以ZORBAX Eclipse XDB-C18(250mm×4.6mm,5μm)为色谱柱;流动相为乙腈和水,梯度洗脱,流速1mL·min-1,柱温35℃,蒸发光散射检测器,ELSD参数:喷雾器级别是66%,漂移管温度55℃,载气压力25psi,增益值为200。结果:用梯度洗脱得到的色谱图各色谱峰分离较好,达到指纹图谱要求。用主成分分析和聚类分析法把14个不同产地的黄芪样品分为5类。结论:为更好的控制黄芪的内在质量提供了可靠的分析方法。

Abstract:Objective: To establish a fingerprint analysis method of Radix Astragali from various habitats. Methods: The fingerp rint was performed by HPLC-ELSD. The conditions of chromatography: ZORBAX Eclipse XDB-C18( 250mm × 4. 6 mm,5 μm ) was used with a mobile phase of CH3CN-H2O,flow rate:1mL·min-1,column temperature: 35℃,neb: 66% ,tube: 55℃,air pressure: 25psi,gain:200. Results: HPLC fingerprint of Radix Astragali was obtained. The 14 samples were classified as 5 clusters by PCA and clusteranalysis. Conclusion:A reliable method was provided for controlling the quality of Radix Astragali.
