白藜芦醇诱导人宫颈癌Hela细胞凋亡及其机制

摘    要:目的探讨白藜芦醇诱导人宫颈癌Hela细胞的凋亡作用及其分子机制。方法应用不同浓度的白藜芦醇作用于人宫颈癌Hela细胞,采用四甲基偶氮唑蓝(MTT)法检测药物对细胞的增殖抑制率;流式细胞术(FCM)检测细胞凋亡、细胞周期分布:免疫组化法检测Survivin及Caspase-3的表达。结果白藜芦醇能明显抑制Hela细胞的增殖,并呈剂量和时间依赖性。经白藜芦醇处理Hela细胞后,FCM分析发现各实验组S期细胞比例增高,G2/M期细胞比例减少,凋亡率明显高于对照组,并呈剂量依赖性(P〈0.01);各实验组Heal细胞Survivin的表达均低于对照组,而Caspase-3的表达均高于对照组(P〈0.01)。结论白藜芦醇能明显抑制人宫颈癌Hela细胞的增殖,诱导其凋亡,其机制可能与抑制Survivin的表达、上调Caspase-3的表达有关。

Mechanism of the apoptosis induced by resveratrol in human cervical cancer Hela cells

Abstract:Objective To investigate human cervical cancer Hela cell apoptosis induced by resvertrol and its molecular mechanism. Methods Hela cells were treated with resveratrol of different concentrations.MTT method was used to detect the inhibition rate.Flow cytometry was used to detect the proliferation and apoptosis of the cells.The expression of survivin and caspase-3 of Hela cells were measured with immunohistochemical approach. Results The cell growth was inhibited significantly,with a time and dose dependent fashion.After resveratrol treatment, the apoptosis of Hela cells were induced.FCM analysis found that the number of treated cells in S phase increased, but that in G2/M phase decreased. The apoptosis rate was significantly higher than that of control group ,with a dose dependent fashion (P〈0.01); The expression survivin in treated cells were lower than that of control group, but the expression caspase-3 in treated cells were higher than that of control group (P〈0.01). Conclusion Resveratrol can significantly inhibit proliferation and induce apoptosis of Hela cells.Its molecular mechanisms might be related to modulation the expression of surviving and caspase-3.
