对动物组织样品中砷的检测方法研究

目的：考察不同的样品预处理方法对测定动物组织样品中砷含量的影响,并建立定性定量检测动物组织样品中砷含量的分析方法。方法：分别通过消化法与灰化法处理动物组织的样品,借助二乙基二硫代氨基甲酸银分光光度法Ag（DDC）法]进行砷含量的检测,并且比较其检测结果;采用铜片法对砷进行快速定性检验。结果：Ag（DDC）法所得吸光度对应砷含量的标准曲线在0.00-4.8 mg/L范围内线性良好,相关系数r为0.998 9;砷的检出限为40.6μg/L。两种预处理方法对检测样品的结果影响无明显差异（P〉0.05）。对于不同的动物组织,胃中的砷检出量最大,占给药量的50%;动物组织样品中采用铜片法的检出限约7.5 mg/L。结论：Ag（DDC）法和铜片法分别适用于动物组织样品中砷元素的定量和定性检测;消化法或灰化法均可适用于动物组织样品的预处理;在对大鼠胃、肝、肠、肾等组织样品进行砷含量检测中,胃是最佳检测部位。

STUDY ON VARIOUS METHODS FOR DETERMINING THE ARSENIC IN ANIMAL TISSUES

Objective：To study the influence of different sample pretreatment methods on the determination of the arsenic content in animal tissues,and to build up analytical methods for qualitatively and quantitatively detecting the arsenic content.Methods：The animal tissue samples were pretreated by digestion method and ash method,respectively.The content of arsenic was determined by the silver diethyl dithiocarbamate spectrophotometry Ag（DDC） method] and the results were compared with each other.Copper method was employed to rapidly and qualitatively test the arsenic.Results：The standard absorbance-arsenic content curve obtained by Ag（DDC） method had good linearity in the range of 0.00～4.8 mg/L.The linear correlation coefficient R was 0.998 9,and the detection limit for arsenic was 40.6 μg /L.These two pretreatment methods made no difference to the detecting results（P〉0.05）.Among different animal tissues,the arsenic content detected in the stomach was the highest,which reached 50% of the administering dose.By using copper testing method,the detecting limit of arsenic in animal organs was approximately 7.5 mg/L.Conclusion：Ag（DDC） method and copper method are effective in quantitative and qualitative detection of arsenic in animal tissues,respectively.Both digestion and ash methods can be used to pretreat the animal samples.Among the rat tissues such as stomach,liver,intestine and kidney,the stomach is the best one for quantitatively detecting the arsenic.
