糖脂消对胰岛素抵抗性高血压大鼠胰岛素抵抗性和血清瘦素水平的影响

摘    要:目的 观察糖脂消对胰岛素抵抗性高血压大鼠胰岛素抵抗性和血清瘦素水平的影响。方法 喂雄性 SD大鼠高果糖饲料造成胰岛素抵抗高血压大鼠模型 ,然后随机将其分为 3组 ,分别喂高果糖饲料 ,高果糖加低、高剂量糖脂消 4周 ,观察各组收缩压 (SBP)、空腹血糖 (FBG)、空腹血清胰岛素 (FINS)和瘦素水平 (L P) ,用稳态模型 (HOMA)评估胰岛素抵抗 (IR) ,并对检测的指标进行相关性分析。结果  1与对照组相比 ,高果糖饲料组 SBP、FINS、HOMA- IR明显升高(P<0 .0 1,P<0 .0 1,P<0 .0 5 ) ,FBG无明显变化 (P>0 .0 5 ) ;L P显著升高 (P<0 .0 5 )。2高果糖饲料组大鼠 ,L P与 FINS(r=0 .6 6 6 )、SBP显著正相关 (r=0 .6 82 ) ,与 HOMA- RA显著负相关 (r=- 0 .6 77) ,P值均小于 0 .0 5 ,有显著意义。 3与模型对照组比较 ,高剂量糖脂消组大鼠 HOMA- IR、FINS和 L P显著降低 (P均 <0 .0 5 )。结论 高果糖饲料喂养 SD大鼠能形成胰岛素抵抗及高血压 ,并有高瘦素血症。糖脂消能有效改善胰岛素抵抗 ,糖脂消改善胰岛素抵抗的部分机理可能与其降低血清瘦素水平有关。

Abstract:Objective To observe the effects of Tangzhixiao on insulin resistance and leptin level in high fructose-fed rats.Methods Male Spraque-Darley rats were fed on high fructose diet to establish insulin resistant hypertensive rat model and then randomly divided into three groups.The rats were given high fructose diet or high fructose diet containing two doses of TangZhixiao for 4 weeks. The concentrations of serum glucose,insulin and leptin were determined. The HOMA model was used to evaluate insulin resistance(IR).The relationships between each parameter were analyzed.Results (1)SBP,insulin level and HOMA-IR in rats fed on high fructose were significant higher than those in normal control group,following hyperleptinemia(P<0.05 or P<0.01).(2)In rats fed on high fructose,leptin was positively related to SBP and insulin ,while negatively to HOMA-IR (all P<0.05).(3)The insulin,leptin and HOMA-IR of high dose of Tangzhixiao group were decreased than those in model control group(all P<0.05).Conclusion High fructose diet can induce insulin resistance,hypertension and hyperleptinemia in the SD rats.Tangzhixiao can effectively prevent insulin resistance.The mechanism of Tangzhixiao preventing insulin resistance may be obtained by decreasing leptin level.
