心炎宁颗粒剂对柯萨奇B_3病毒所致病毒性心肌炎小鼠心肌病理改变和超微结构的影响

摘    要:目的:探讨心炎宁颗粒剂对柯萨奇B_3病毒性心肌炎(VMC)小鼠心肌病理改变的影响。方法:Balb/c小鼠腹腔注射柯萨奇B_3病毒感染建立VMC小鼠模型,予心炎宁颗粒剂(高、中、低剂量)灌胃治疗,于不同时间观察心肌病理及超微结构改变,计算心肌病理组织学积分。结果:第3天,模型组和治疗给药组小鼠开始出现活动减少等表现,心肌出现不同程度的病理改变,病理积分有不同程度的升高。第5天时,模型组和低剂量组小鼠一般状况差,开始出现死亡,心肌病理改变加重,病理积分明显升高;而高、中剂量组小鼠一般状况好于上述两组,心肌病理改变轻微,病理积分轻度升高。第7天,模型组小鼠出现死亡高峰,而治疗给药组小鼠开始出现死亡,病理积分开始下降。至第14天,模型组心肌病理积分轻度下降,治疗给药组与模型组比较病理积分下降明显,尤其是高剂量组,差异有统计学意义(P〈0.01)。在整个观察过程中,模型组心肌超微结构改变重于治疗给药组,而低剂量组重于高、中剂量组。结论:心炎宁颗粒剂可以改善发病小鼠的一般状况,减少动物死亡数目,能有效地维护其心肌纤维及膜系统的稳定性。

Abstract:Objective:To study the influence of Xinyanning granules on myocardial pathological changes of viral myocarditis(VMC)mice caused by coxsackie B_3.Methods:Balb/c intraperitoneal injection of B_3 coxsackie virus was used to establish VMC model.Xinyanning granules of high,medium and low doses were given for irrigation;at different times the myocardial ultrastructure and pathological changes were observed to calculate myocardial histologic points.Results:On the 3rd day,the mice of model and treatment groups began to reduce activities;different degrees of myocardial pathological changes appear.On the 5th day,mice of model group and low dose group were in poor conditions and start to die;heart pathological changes were aggravating;pathological integral increased significantly.But mice of high and medium dose group were in good condition,myocardial pathological changed slightly,pathological integral moderately elevated.On the 7th day,mice of model group appear death peak,pathological integral began to decrease.On the 14th day,myocardial pathological integral of model group mildly declined,compared with model group,pathological integral of treatment group decreased significantly,especially in the high-dose group,the difference was statistically significant(P0.01).In the whole process of observation,myocardial ultrastructures changes of model group were more severe than that of treatment group.Conclusion:Xinyanning granules can improve the general conditions of mice,reduce the number of dead animals,and effectively maintain the myocardial fibers and membrane system stability.
