新疆阿魏蘑菇提取物抗肿瘤实验研究

摘    要:目的 : 探讨新疆阿魏蘑菇提取物对体外培养的肿瘤细胞生长的抑制作用及其有效成分。方法 : 采用肿瘤细胞体外培养技术 ,观察阿魏蘑菇不同剂量的水提物和醇提物 ( 1 0 -1、1 0 -2 、1 0 -3 、1 0 -4、1 0 -5、1 0 -6kg/L)对体外培养的人肝癌细胞株 ( Q3 )、人胃癌细胞株 ( MGC- 80 3)、人宫颈癌细胞株 ( Hela)、小鼠肺腺瘤细胞株 ( SPC- A- 1 )细胞存活及蛋白合成的影响。以大鼠肝细胞( BRL)为正常对照细胞株 ,同时设立空白对照、阴性对照及溶剂对照。受试物与细胞共培养 2 4 h,观察细胞存活量及活细胞蛋白含量的变化 ;采用系统定性测试方法 ,对阿魏蘑菇中可能存在的抗肿瘤有效成分进行测定。结果 : 阿魏蘑菇水提物、醇提物 1 0 -1kg/L剂量对各类型肿瘤细胞的生长及蛋白合成均有明显抑制作用 ;醇提物不同剂量组对肿瘤细胞生长及蛋白合成可产生不同程度的影响 ,对正常肝细胞的存活量无影响 ,且可促进肝细胞的蛋白合成。阿魏蘑菇中可能有糖类、皂甙、蛋白质、多肽、氨基酸、有机酸、生物碱、挥发油、三萜或甾醇、带双键的萜类等有效成分。结论 : 阿魏蘑菇提取物的抗肿瘤作用可能与其抑制肿瘤细胞生长及蛋白合成有关 ,阿魏蘑菇中确切的功效成分及其与抗肿瘤作用之间的关系有待进一步探讨。

Abstract:Objective: The effect of Pleurotas sapidus on 4 kinds of tumor cell lines(Q 3,MGC 803, Hela, SPC A 1) and the effective components were studied.Methods: Four tumor cell lines were cultured in vitro. Normal rat liver cell line served as control. We investigated the effect or water extract or ethanol extract from Pleurotas sapidus with doses 10 -1 -10 -6 kg/L, dissolvent control being normal saline and 7.5% glycerine. A blank control was set up in each group. The survival rate and the protein content of the cultured cells were observed at 24 h culture. We also analyzed the effective components in Pleurotas sapidus. Results: (1)There was a significant growth inhibition on four carcinoma cell lines by 10 -1 kg/L dose of water extract and ethanol extract; (2)The growth inhibition on cultured cells by ethanol extract in different doses were different. No inhibited effect on normal liver cells was found. In Pleurotas sapidus the antitumorous components perhaps were polysaccharide, polypeptide, amino acid, organic acid and alkaloid etc. Conclusion: Extracts from Pleurotas sapidus can inhibit the growth of carcinoma cell lines, probably by inhibiting cell growth and protein synthesis.
