股骨头坏死愈胶囊对兔激素性股骨头坏死血管内皮生长因子表达影响的实验研究

目的观察股骨头坏死愈胶囊对兔激素性股骨头缺血性坏死血管内皮生长因子(VEGF)表达的影响,探讨其对激素性股骨头坏死(SANFH)的防治效果及作用机制。方法将诱导成功的模型分为正常组,模型组,通络生骨胶囊组,股骨头坏死愈胶囊低剂量组、高剂量组,分别给予通络生骨胶囊、股骨头坏死愈胶囊混悬液灌胃,于治疗8周后处死取材,行常规HE染色及VEGF染色观察。结果正常组VEGF表达的阳性细胞数目比率最高,模型组最低,股骨头坏死愈胶囊高剂量组和通络生骨胶囊组相当,但较正常组均低,且高剂量组和通络生骨胶囊组均高于低剂量组(P<0.05)。结论股骨头坏死愈胶囊抑制了激素促使VEGF表达持续减弱的趋势,有效改善股骨头内微循环,扭转股骨头缺血状态,促进骨小梁再生与修复,达到防治SANFH的发生和发展。

Experimental Research of the Impacts on VEGF for the Rabbits with Steroid-Induced Femoral Head Necrosis Treated with Gugutouhuaisiyu Capsules

Objective To observe the impacts on vascular endothelial growth factor（VEGF）in the rabbits with steroid-induced avascular necrosis of femoral head（SANFH）treated with gugutouhuaisiyu capsules and explore the effect and the mechanism of the capsules on the prevention and treatment of SANFH.Methods The successfully induced models were divided into a normal group,a model group,a tongluo zhenggu capsules group and the low-dose and high-dose gugutouhuaisiyu capsules groups.The lavage of the suspensions of tongluo zhenggu capsules and gugutouhuaisiyu capsules was administrated separately.The rabbits were sacrificed for the material collection in 8 weeks of treatment.The routine HE staining and VEGF staining were adopted for observation.Results In the normal group,the positive cell count proportion of VEGF expression was the highest and that in the model group was the lowest.The result in the high-dose gugutouhuaisiyu capsules group was same as that in the tongluo shenggu capsules group,but the result in either group was lower than that in the normal group.The result in either the high-dose gugutouhuaisiyu capsules group or the tongluo shenggu capsules group was higher than that in the low-dose gugutouhuaisiyu capsules group（P0.05）.Conclusion Gugutouhuaisiyu capsules inhibit the steroid-induced persistent weakening tendency of VEGF expression.This drug effectively improves the inner microcirculation of femoral head,reverses the avascular state of femoral head and promotes the regeneration and repair of trabecular bone so as to prevent from the occurrence and development of SANFH.
