摘    要:
目的:比较附子不同的样品制备方法,优化附子中3种生物碱的分离条件。方法:采用反相高效液相色谱法测定附子中生物碱的含量,色谱柱为Zorbax extend-C1(8150mm×4.6mm,5μm)。比较采用不同方法制备的样品中生物碱含量的差异。结果:乙醚超声提取的方法最优;分离条件:流动相为乙腈-0.1%乙二胺,梯度洗脱,流速为lmL·min-1,检测波长为240nm,柱温为30℃。结论:乙醚超声提取方法过程简单、分离效果好、提取率和回收率高,可为附子的质量控制和成分研究提供科学依据。

Abstract:
OBJECTIVE:To compare the different sample preparation and optimize the isolation of three kinds of alkaloids in Aconitum carmichaeli.METHODS:RP-HPLC was applied and performed on Zorbax extend-C18 column (150 mm×4.6 mm,5 μm) column.The differences among the content of alkaloids prepared by preparation technologies were compared.RESULTS:The best preparation was aether ultrasonic extraction.The separation condition was as follows:mobile phase consisted of acetonitrile-0.1% ethylenediamine (gradient elution) with flow rate of 1 mL·min^-1.The detection wavelength was set at 240 nm and column temperature was 30 ℃.CONCLUSION:Aether ultrasonic extraction is simple with ideal separation,extraction and re-covery to provide scientific basis for the quality control of A.carmichaeli.
