黄连素对PCOS大鼠胰岛素抵抗通路关键蛋白的影响

摘    要:目的通过观察实验动物检测指标变化,探究卵巢局部胰岛素信号通路在PCOS形成过程中的作用,并评价黄连素的治疗作用。方法选用清洁级SD雌性大鼠,予以硫酸普拉睾酮钠颈背部皮下注射建立PCOS大鼠模型,并予黄连素干预,对照组选用二甲双胍干预。干预疗程结束后检测各组大鼠空腹血糖(FPG)、口服葡萄糖耐量试验(OGTT)及空腹胰岛素(FINS)水平,计算葡萄糖曲线下面积及稳态模型胰岛素抵抗指数(HOMA-IR)。利用WB技术检测各组大鼠卵巢组织中PI3K/Akt/GSK-3β信号通路中关键蛋白的表达情况。结果各治疗组大鼠OGTT试验给糖后30 min和60 min血糖水平降低、葡萄糖曲线下面积减少、FINS水平、HOMA-IR指数降低,具有统计学意义(P<0.05)。与模型对照组大鼠相比,黄连素治疗组PCOS模型大鼠卵巢组织PI3k P85α、Akt2、p GSK-3β(Thy216)蛋白表达升高;p GSK-3β(Ser9)表达降低,差异显著(P<0.05),具有统计学意义。结论黄连素能够改善PCOS大鼠血清及卵巢局部胰岛素抵抗,其可能机制为良性调节PI3K/Akt/GSK-3β胰岛素信号通路中关键蛋白表达,这可能是黄连素治疗PCOS的作用机制之一。

Abstract:Objective To investigate the role of ovarian local insulin signaling pathway in the formation of PCOS by observing changes in experimental animals, and to evaluate the therapeutic effects of berberine.Methods Clean-grade SD female rats were used to establish a PCOS rat model by subcutaneous injection of prasugrel sulfate sodium in the back of the neck. The PCOS rat model were treated with berberine, and metformin was used in the control group.At the end of the intervention period, fasting blood glucose(FPG), oral glucose tolerance test(OGTT) and fasting insulin(FINS) levels were measured in each group, and the area under the glucose curve and the homeostasis model insulin resistance index(HOMA-IR) were calculated. WB technique was used to detect the expression of key proteins in PI3 K/Akt/GSK-3β signaling pathway in ovarian tissues of rats.Results The blood glucose levels of the OGTT rats in each treatment group were decreased 30 min and 60 min after glucose administration, the area under the glucose curve decreased, the FINS level and the HOMA-IR index decreased, which was statistically significant(P < 0.05). Compared with the model control group, the expression of PI3 k P85α, Akt2 and pGSK-3β(Thy216) protein in ovarian tissue of berberine treatment group was increased,and the expression of pGSK-3β(ser9) was decreased, the difference was significant(P < 0.05), which has statistical significane. Conclusion Berberine can improve the serum and ovarian insulin resistance of PCOS rats. The possible mechanism is the benign regulation of key protein expression in PI3 K/Akt/GSK-3β insulin signaling pathway, which may be one of the mechanisms of berberine treatment of PCOS.
