复方三金胶囊对乳腺癌荷瘤裸鼠瘤组织细胞增殖的影响

目的研究复方三金胶囊对乳腺癌MCF-7细胞裸鼠体内的细胞抑制作用及其作用机制。方法将30只裸鼠接种人乳腺癌MCF-7细胞,待瘤体积超过5 mm2时,将30只裸鼠随机分为模型组、复方三金胶囊高、中、低剂量组及环磷酰胺对照组,连续给药2周后,取血、瘤组织及脾脏,用于各项指标测定。通过测量给药前后荷瘤小鼠瘤的重量,计算出复方三金胶囊各剂量组的抑瘤率;待裸鼠瘤体积超过5 mm2时,每3 d测量1次裸鼠瘤的长径与短径,用于计算瘤体积,观察复方三金胶囊对小鼠瘤体积的影响。结果在瘤体积方面,与模型组比较,复方三金胶囊高、中、低剂量组于给药第14天、17天、20天、23天瘤体积均降低(P <0.01),给药第26天,复方三金胶囊高、中剂量组瘤体积降低(P <0.05),环磷酰胺组各时间段瘤体积均降低(P <0.01);在瘤重方面,与模型组比较,复方三金胶囊高、中剂量组、环磷酰胺组均可以显著降低瘤重(P <0.01);在抑瘤率方面,复方三金胶囊高、中、低剂量组的抑瘤率分别为44.76%、33.12%、14.61%,环磷酰胺对照组的抑瘤率为43%;在脾重及脾脏指数方面,与模型组比较,复方三金胶囊高剂量组能够显著增加脾重、增大脾脏指数(P <0.01),低剂量组能够降低脾脏指数(P <0.01)。结论复方三金胶囊能够显著抑制乳腺癌细胞的增殖,表现在能够显著抑制肿瘤体积的增长、降低瘤重及对脾脏无明显影响。

Effect of Fufang Sanjin Capsule on Proliferation of Tumor Tissue in Nude Mice with Breast Cancer

Objective To study the inhibitory effect of Fufang Sanjin capsule on the cytostatic effect of breast cancer MCF-7 cells in nude mice and its mechanism. Methods Thirty nude mice were inoculated into human breast cancer MCF-7 cells. When the tumor volume exceeded 5 mm^2, 30 nude mice were randomly divided into model group, Fufang Sanjin capsule high, medium and low dose groups and cyclophosphamide control group. In the group, after 2 weeks of continuous administration, blood, tumor tissue and spleen were taken for determination of various indexes. The tumor inhibition rate of Fufang Sanjin capsule was calculated by measuring the weight of tumor-bearing mice before and after administration. When the tumor volume of nude mice exceeded 5 mm^2, the long and short diameters of nude mice were measured every 3 days. For the calculation of tumor volume, observe the eff ect of Fufang Sanjin capsule on tumor volume in mice. Results In terms of tumor volume, compared with the model group, the tumor volume of Fufang Sanjin capsule in the high, middle and low dose groups decreased on the 14 d, 17 d, 20 d and 23 d of administration(P < 0.01). On the 26 d after administration, the high and medium doses of Fufang Sanjin capsule could reduce the tumor volume(P < 0.05), and the tumor volume decreased in each period of the cyclophosphamide group(P < 0.01). In terms of tumor weight, compared with the model group, the Fufang Sanjin capsule high, middle dose group, cyclophosphamide group can significantly reduce the tumor weight(P < 0.01). In terms of tumor inhibition rate, the inhibition rates of Fufang Sanjin capsule in the high, middle and low dose groups were 44.76%, 33.12% and 14.61%, respectively, and the inhibition rate of the cyclophosphamide control group was 43%;in the spleen weight and spleen index, compared with the model group, the Fufang Sanjin capsule high-dose group can significantly increase the spleen weight and increase the spleen index, which is extremely significant(P < 0.01). The Fufang Sanjin capsule low-dose group can reduce the spleen index(P < 0.01). Conclusion Fufang Sanjin capsule can significantly inhibit the proliferation of breast cancer cells, which can significantly inhibit the growth of tumor volume and have no obvious eff ect on the spleen.
