三氧化二砷联合PDTC诱导胃癌细胞凋亡及其对VEGF表达的影响

摘    要:目的 研究三氧化二砷(arsenic trioxide,As2O3)和吡咯烷二硫代氨基甲酸盐(pyrrolidine dithiocarbamate,PDTC)联合诱导胃癌细胞.SGC-7901凋亡及对血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响. 方法 用不同浓度的As2O3和(或)PDTC处理SGC-7901细胞,用四甲基偶氮唑蓝(MTT)法测细胞生长抑制率,用流式细胞仪(ECM)检测细胞周期及细胞凋亡,并用免疫细胞化学方法 检测VEGF的表达. 结果 As2O3可抑制SGC-7901的生长,并随浓度的增加而增强.As2O3与PDTC联合比单用As2O3或PDTC抑制效果更明显(P<0.05).As2O3>作用后SGC-7901 Cn/M期细胞比例上升,联合用药组比单用药组G2/M期细胞比例上升更明显(P<0.05).As2O3可诱导SGC-7901细胞凋亡,联合用药组比单药组组凋亡率增加更明显(P<0.05);As2O3组VEGF表达下降(P<0.05),联合用药组比单药组表达下降更明显(P<0.05). 结论 As2O3与PDTC联合应用具有显著抗胃癌作用,其机制可能与细胞周期阻滞、增强诱导胃癌细胞凋亡以及抑制NF-кB、VEGF表达有关.
