核酸对淋巴细胞DNA损伤修复的影响

摘    要:目的 研究核酸对大鼠淋巴细胞DNA损伤的保护作用。方法 纯种雄性健康Wistar大鼠,随机分为4组:空白组、模型组、核酸低剂量组、核酸高剂量组。空白组正常饮水,其余3组饮0.8%醋酸铅水溶液,核酸组灌胃核酸,空白组、模型组灌蒸馏水,5周后,采用单细胞凝胶电泳技术对外周血淋巴细胞DNA损伤进行分析。结果 模型组大鼠淋巴细胞DNA的迁移率和尾长显著高于空白组,核酸组大鼠淋巴细胞DNA的迁移率和尾长显著低于模型组,在统计学上差异有显著性(P〈0.05)。结论 慢性铅染毒可致大鼠淋巴细胞DNA损伤,饮食核酸对淋巴细胞DNA损伤具有一定的保护作用,可减轻铅中毒引起的氧化损伤。

Abstract:Objective To test the protective effect of dietary nucleic acid on lymphocyte DNA damage repair. (Methods) Inbreed healthy male Wistar rats were experimentally exposed to lead acetate to induce DNA injury and received dietary nucleic acid to test their protective effects on lymphocyte DNA damage repair.After 5 weeks experimental treatment,the DNA breaks in peripheral blood lymphocytes were analyzed with single cell gel electrophoresis. Results Compared(with) control,the DNA damage in dietary nucleic acid supplemented groups were reduced significantly(P<0.05). (Conclusion) (Dietary) nucleic acid could protect DNA damage of peripheral blood lymphocytes in rats.
