Advances in chimeric antigen receptor T cell immunotherapy construction technology

Abstract:Chimeric antigen receptor T cell immunotherapy(CAR-T)is considered to be the most promising treatment method and has achieved good clinical efficacy.With the large-scale clinical application of CAR-T,the limitations of this technology are gradually exposed,such as poor therapeutic effect,disease recurrence and a large number of adverse reactions.This makes the manufacture of CAR-T cells with better structure,higher safety and better efficiency clinically necessary.Understanding the manufacturing process of CAR-T and mastering the technological advances is the key to construct ideal CAR-T cells.This review focuses on CAR-T cell construction technology,especially in the development of chimeric antigen receptor component construction and gene transduction technology,and describes CAR-T cell products in line with the Good Manufactured Practices(GMP).Finally,the development,promotion and application of this technology are prospected.
