多潘立酮片在健康人体的相对生物利用度与生物等效性

目的建立测定血浆中多潘立酮浓度的HPLC—FL法并研究多潘立酮片的相对生物利用度及其生物等效性。方法按两制剂双周期自身对照交叉试验设计，20名男性健康志愿者分别单剂量口服受试制剂和参比制剂多潘立酮片，用HPLC法测定血药浓度，计算药代动力学参数，并评价两制剂的生物等效性。结果口服多潘立酮片（受试制剂）和多潘立酮片（吗丁啉，参比制剂）20mg后的主要药代动力学参数：受试制剂多潘立酮片t1/2（9．14±0．77）h、Cmax（28．94±4．06）ng·mL^-1、Tmax（0．75±0．24）h、AUC0-1（144．36±30．94）ng·h·mL^-1；参比制剂多潘立酮片t1／2（8．58±1．10）h、Cmax（29．62±5．01）ng·mL^-1、Tmax（0．79±0．32）h、AUC0-1（140．51±31．13）ng·h·mL^-1。以AUC0-1计算，与参比制剂相比受试制剂中多潘立酮的平均相对生物利用度为（103．0±5．4）％。结论两制剂具有生物等效性。

Relative Bioavailability and Bioequivalence of Domperidone Tablets in Healthy Volunteers

Aim To determine domperidone concentrations in human plasma and study the pharmacokinetics and relative bioavailability of domperidone tablets. Methods A single dose of 20mg of reference and test domperidone tablets were given to 20 healthy volunteers in a randomized 2-way cross-over study. The plasma concentrations of domperidone were determined by HPLC-FL. The pharmacokinetic parameters as well as relative bioavailability were measured. Results The main pharmacokinetic parameters were t 1/2 （ 9. 14 ± 0.77 ） h, C max （ 28.94 ± 4.06 ） ng·mL^-1 , T max （0.75 ± 0. 24） h, AUC0-1 （ 144.36 ± 30.94） ng·h·mL^-1 for the test drug and t 1/2 （ 8.58 ± 1.10 ） h, Cmax（29. 62 ± 5. 01） ng·mL^-1, Tmax（0.79 ±0.32）h,AUC0-1（140.51 ±31.13）ng·h·mL^-1 for the reference drug. The relative bioavailability of AUC0-30 was （103.0 ± 5.4）%. Conclusion The reference and test domperidone tablets are bioequivalent.
