OBSERVATION ON CHRONiC TOXICITY OF α-CHLOROHYDRIN COMBINED WITH GLYCIDOL IN MONKEYS (MACACA MULATTA) |

Abstract: | Five adult monkeys were orally admistered with a-chlorohydrin combined with glycidol in six consecutive courses of treatment, each of which included two-days-admistration with a daily dose of 5mg/kg a-chlorohydrin and 75mg/kg glycidol and eight-days-cessation of medication. Three untreated monkeys were used as controls. For histological examination, the left testis and epididymis were extirpated at the end of 3rd course of treatment; the right testis and epididymis, bonemarrow and other inner organs were taken from the sacrificed monkeys at the end of 6th course of treatment. Additionally blood cell count, differential blood cell count, hemoglobinmetering and chromosome study were done at two intervals mentioned above. Damage to the spermatogenesis and sperm blockage of ductus epididymis were not recognized. The changes of blood cell count and differential cell count, and hemoglobin content were not significant. No histo-pathological changes standing for a chronic damage in inner organ could be found. No significant chromosome aberration was noted. Almost all of the monkeys had vomiting one and a half hours or three hours after the first administion of each course of treatment. The results suggest that a low combined dose of α-chlorohydrin and glycidol used twice every 10 days for a period of 60 days in monkeys has no significant toxicologic effects, although the monkeys do have vomiting. |
