高同型半胱氨酸血症对血脑屏障的影响

摘    要:目的建立高同型半胱氨酸血症大鼠模型,观察高同型半胱氨酸血症对血脑屏障的影响。方法36只SD大鼠分为对照和模型(皮下注射蛋氨酸)两组。采用高压液相色谱法检测同型半胱氨酸(homocysteine,Hcy)浓度;比色法检测大鼠大脑伊文思蓝含量观测血脑屏障的通透性改变;透射电镜观察大脑皮质毛细血管形态改变。结果对照组大鼠在不同时间点血浆Hcy水平和脑组织伊文思蓝含量均无明显变化,血脑屏障结构亦正常。模型组大鼠血浆Hcy水平和脑组织伊文思蓝含量在第4w和第8w均显著增高,同时血脑屏障也呈现不同程度的破坏。结论高同型半胱氨酸血症可损伤大鼠血脑屏障的结构和功能。

Abstract:Objective To establish hyperhomocysteinemia (HHcy) rat model and to observe the effect of HHcy on boold-brain barrier. Method Thirty-six rats were randomly divided into two groups: control group and model group. Model group was given dl-methionine (80 mg/kg?d) subcutaneously. The concentration of plasma homocysteine (Hcy) was measured with high-performance liquid chromatography. To observe the change in permeability of the boold-brain barrier, Evans blue in the brain tissue was surveyed by colorimetry. The histological changes of the capillary vessel were examined under electron microscope. Results In control group, no differences were observed in level of plasma Hcy , contents of Evans Blue and the structure of the boold-brain barrie. Level of plasma Hcy and the contents of Evans Blue of model group was significantly higher than those of control group at 4 w and 8 w after injection. At the same time, the different breakdown of the boold-brain barrie appeared under electron microscope in model group. Conclusion Elevated levels of plasma hyperhomocysteinemia could damage the structure and function of the blood-brain barrier in rats.
