肥胖大鼠心肌细胞内游离钙浓度的变化

目的探讨肥胖大鼠心肌细胞内钙调节机制。方法 60只健康雄性SD大鼠高脂饲料喂饲10周后,筛选出肥胖易感(OP)大鼠,基础组大鼠和肥胖抵抗(OR)大鼠,每组15只。测量各组大鼠体脂肪含量,检测血脂水平。采用酶法分离心肌细胞,Fluo-3/AM负载后采用激光共聚焦扫描显微镜观察各组大鼠心肌[Ca2+]i对KCl除极及咖啡因诱导的反性。结果与基础组大鼠和OR大鼠比较,OP大鼠睾周脂肪、肾周脂肪、网膜脂肪、体脂比升高(P<0.05),血清总胆固醇、三酰甘油和低密度脂蛋白水平升高(P<0.05),心室肌[Ca2+]i的升高幅度降低(P<0.05)。结论肥胖心肌细胞[Ca2+]i在KCl除极后及咖啡因诱导后升高幅度发生改变,可能是肥胖者心律失常发生机制之一。

Change of Intracellular Calcium in Myocardial Cell of Obesity Prone Rats
