高原低氧对健康人体内磺胺甲噁唑的蛋白和红细胞结合率的影响

摘    要:目的观测高原低氧对健康人体内磺胺甲噁唑的蛋白和红细胞结合率的影响。方法取平原和高原地区健康男性志愿者口服磺胺甲噁唑后1.5、12和48h时间点血样,抗凝,一半保留全血,另一半分离血浆,超率法测定蛋白结合率,红细胞结合率用红细胞压积、血浆和全血药物浓度计算。药物浓度均采用反相高效液相色谱法测定。结果平原健康男性志愿者体内磺胺甲噁唑的蛋白和红细胞结合率分别为65.31%和5.96%,高原健康男性志愿者体内磺胺甲噁唑的蛋白和红细胞结合率分别为69.60%和9.41%,高原健康男性志愿者体内磺胺甲噁唑的蛋白和红细胞结合率显著高于平原健康男性志愿者。结论高原低氧使健康人体内磺胺甲噁唑的蛋白和红细胞结合率均显著升高。

Abstract:Objective To study the effect of high altitude hypoxia on the binding of protein and red blood cells of sulfamethoxazole in healthy volunteers.Methods 1.5、12 and 48 h after volunteers living at plain and high altitude were administrated orally cotrimoxazole tablets,the blood sample was collected in order to obtain whole blood and plasma samples.Protein binding was determined by ultrafiltration method,and RBC binding was calculated with hematocrit,whole blood and plasma concentration.The concentration of sulfamethoxazole in biological sample was determined by RP-HPLC method.Results The protein and RBC binding of sulfamethoxazole in healthy volunteers living at plain were 65.31% and 5.96%,respectively,and The protein and RBC binding of sulfamethoxazole in healthy volunteers living at high altitude were 69.60% and 9.41%,respectively.The protein and RBC binding of sulfamethoxazole in healthy volunteers living at high altitude were significantly higher than healthy volunteers living at plain.Conclusion The effect of high altitude hypoxia on the protein and RBC binding of sulfamethoxazole in healthy volunteers was significant.
