摘    要:目的比较研究具潜在药用价值的亚心形扁藻Platymonas subcordiformis、眼点拟微绿球藻Nannochloropsis ocutala、海洋小球藻Chlorella vulgaris 3种绿藻门海洋微藻的细胞生长特性,为海洋微藻在医药保健食品行业的应用提供参考依据。方法选取上述3种海洋微藻为生物材料,以添加f/2营养液的人工海水为培养介质,在光照培养箱中利用三角瓶开展一次性培养实验。利用显微镜观测微藻生长14 d内的细胞密度变化情况,采用最小二乘法拟合细胞密度法计算微藻生长速率。结果在实验周期内,3种海洋微藻的细胞生长曲线均呈现出"S"型。在生长平稳期,亚心型扁藻、眼点拟微绿球藻和海洋小球藻的细胞密度分别约为1.30×106、3.50×107和2.45×107个/mL;最大生长速率分别约为0.7/d、0.8/d和1.5/d。结论上述3种具药用价值绿藻的细胞增长经历明显的缓慢期、快速期和平稳期阶段;不同微藻的细胞生长特性差异较大,在进行其高密度培养开发应用时应结合考虑品种特性与收获时期。

Abstract:Objective To study comparatively on the cell growth characteristics of three marine microalgae,including Platymonas subcordiformis,Nannochloropsis ocutala and Chlorella vulgaris in Chlorophyta with medicinal potential,so as to provide useful information for the further application of marine microalgae.Methods The microalgae were cultured in plant growth charmer in f/2 enriched artificial sea water with conical flasks.Cell numbers were counted with a microscopy,and daily growth rate was calculated by least square method for 14 days.Results Three microalgae showed S-shaped growth curves during the experimental period.In the stable growth stage,the cell densities were 1.30×106,3.50×107 and 2.45×107 cells/mL,and their maximal growth rates were 0.7,0.8 and 1.5/d,respectively,for Platymonas subcordiformis,Nannochloropsis ocutala and Chlorella vulgaris.Conclusion The three microalgae experience the slow,rapid and stable growth stage during the experiment.The growth characteristic varied greatly with different microalgae,hence it is necessary to consider both species′ feature and growth stage when developing high-density culture for medicinal purpose.
