红景天苷对百草枯中毒大鼠肺组织保护作用的研究

摘    要:目的 通过观察红景天苷对百草枯中毒大鼠肺组织转化生长因子-β 1(TGF-β 1)表达、TNF-α含量和血清IL-6水平的影响,探索其对肺损伤的保护作用.方法 将90只SD大鼠随机分为正常对照组、百草枯模型组、红景天苷治疗组,后两组再分为1、6、24、72h 4个亚组,每组10只.百草枯模型组和红景天苷治疗组采用百草枯溶液1ml/kg一次性灌胃制备急性百草枯中毒肺损伤模型,然后分别予等容积的0 9%氯化钠溶液、红景天苷1m l/kg腹腔注射.3组大鼠麻醉后,取股动脉血测定IL-6水平,处死后取肺组织,观察病理变化,采用免疫组化和RT-PCR测定肺组织中TGF-β 1及其mRNA的表达情况,并测定肺组织中TNF-α含量.结果 百草枯模型组大鼠肺组织出现大片出血及坏死,红景天苷治疗组大鼠肺组织损伤程度较百草枯模型组减轻.与正常对照组比较,百草枯模型组、红景天苷治疗组肺组织6、24、72h TGF-β 1表达显著增加(均P<0.01),但红景天苷治疗组较百草枯模型组为低(均P<0.05).百草枯模型组与红景天苷治疗组中毒后第1h起大鼠肺组织TGF-β 1mRNA水平开始增加,但红景天苷治疗组较百草枯模型组为低(均P<0.05).与正常对照组比较,百草枯模型组及红景天苷治疗组肺组织中TNF-α含量、血清IL-6水平从第6h开始显著增加(均P<0.01),但红景天苷治疗组在各时点均较百草枯模型组为低(均P<0.05).结论 TGF-β1参与了百草枯中毒大鼠肺损伤的早期病理生理过程,红景天苷能抑制TGF-β 1的表达,降低肺组织TNF-α含量、血清IL-6水平,从而减轻百草枯对大鼠肺组织的损伤.

Protective effect of salidroside on paraquat poisoned rat lung tissue and its mechanism

Abstract:Objective To investigate the effect of salidroside (SDS) on paraquat poisoned rat lung tissue and its mechanism. Methods Ninety Sprague-Dawley (SD) rats were randomly divided into normal control group (n=10), paraquat model group (n=40) and SDS treatment group (n=40). Rats in model and treatment groups were administered with a single dose of PQ (1ml/kg) by gavage; then lml/kg salidroside (treatment group) or 0.9% saline (model group) was injected intraperitoneally. The rats in model and treatment groups were sacrificed at 1, 6, 24 and 72h after treatment (10 in a batch), the blood samples were taken from femoraL arterial and the lung tissue samples were also collected. The serum IL-6 levels were measured, and the pathological changes in lung tissue were examined; the expression of TGF- β protein and mRNA were detected by immunohistochemistry and RT-PCR, respectively; the TNF-α levels in lung tissue were also measured. Results Severe hemorrhage and necrosis were noticed pathologically in lung tissue of model group, the degree of lung injury in treated group was attenuated compared with model group. Compared with normal control group, the expression on TGF- β 1 in lung issue of both model and treatment groups at 6, 24, 72h increased significantly (all P〈0.01), but the expression in treatment group was lower than that in model group (P〈0.05). The TGF- β 1mRNA level in lung tissue of model and treatment groups began to increase at lh after poisoning, but the level in treatment group was lower than that in model group (P〈0.05). Compared with normal control group, lung tissue TNF-α levels and serum IL-6 levels of model and treatment groups began to increase from 6h after poisoning (P〈0.01), but those in treatment group were lower at each time points (all P〈0.05). Conclusion Salidroside can attenuate the lung tissue injury induced by paraquat in rats, which is associated with down-regulation of TGF-β1, TNF-α expression in lung tissue and decreased
