电针干预高血压大鼠脑缺血血管内皮生长因子和促血管生成素1表达的研究

摘    要:目的观察电针对易脑卒中型肾血管性高血压大鼠(RHRSP)脑缺血再灌注血管内皮生长因子(VEGF)、促血管生成素1(Ang 1)表达及血管新生的影响。方法用环形银夹钳夹SD大鼠的双侧肾动脉,制成RHRSP 120只,随机分为4组:空白组(12只),假手术组(12只)、模型组(48只)和电针组(48只),后2组大鼠再用线栓法制成一侧大脑中动脉闭塞模型,电针组给予"百会"、"大椎"穴位电针治疗,1次/d,14天。免疫组织化学法和光镜等技术观察脑缺血2 h后,再灌注1、7、14、28天大鼠脑内缺血区VEGF、Ang 1表达及缺血区微血管密度(MVD)的变化。结果与空白组和假手术组比较,模型组大鼠7、14、28天梗死灶周围VEGF和Ang 1的表达明显增强(P0.05);与模型组比较,电针组大鼠7、14、28天VEGF和Ang 1的表达均明显增强(P0.05),脑缺血14、28天缺血区及周围MVD明显增加(P0.05)。结论电针对RHRSP脑缺血再灌注损伤的保护作用,可能与增强梗死灶周围VEGF和Ang 1的表达,促进缺血区的血管新生有关。

Effects of electric acupuncture on expression of cerebral ischemic vascular endothelial growth factor and angiopoietin 1 in hypertensive rats
Abstract:Objective To observe the effects of electric acupuncture on the expression of vascular endothelial growth factor,angiopoietin 1 and angiogenesis in stroke-prone renovascular hypertensive rat(RHRSP).Methods 120 RHRSP models were established by bilaterally narrowing the renal arteries with silver loop clips.The RHRSP were randomly divided into 4 groups:naive group,sham-operated group,model group and electric acupuncture group.The models of rat MCAO were created by stringing middle cerebral artery in the two later groups.The electric acupuncture group was treated with electric acupuncture stimulating "BaiHui"and "DaZhui" point once a day for 14 days.VEGF,Ang 1 and microvessel density(MVD)in the rat cerebral ischemic tissue were detected by immunohistochemistry,light microscope,etc.at the different time points after 2 h ischemia and 1,7,14 and 28-day reperfusion.Results The VEGF and Ang 1 positive cells of the ischemic area increased markedly in model group compared with the naive and sham-operated groups (P<0,05).The number of VEGF and Ang 1 positive cells in electric acupuncture group was increased significantly compared with the model group (P <0.05).MVD in electric acupuncture group was markedly increased compared with model group after 14-day and 28- day reperfusion (P < 0.05).Conclusions The electric acupuncture can promote the neurofunctional recovery in cerebral ischemic area.The mechanism may be related with the regulation of the expression of VEGF and Ang 1 and activating angiogenesis.
