黄芪总苷对二甲肼诱导大鼠结直肠异性隐窝灶和肝微粒体代谢酶的影响

摘    要:目的 观察黄芪总苷对二甲肼诱导大鼠产生异性隐窝和肝微粒体细胞色素P450(cytochrome P450 S,CYP450)含量、谷胱甘肽转移酶(glutathione-S-transferase, GST)活力的影响.方法 40只SD大鼠随机分为空白对照组、黄芪组、DMH组、黄芪+DMH组,每组10只.黄芪总甙干预和二甲肼诱癌后脱颈处死动物,取结直肠和肝组织.美蓝染色结直肠后观察、计数ACF的个数,差速离心法分离出大鼠肝微粒体.采用一氧化碳还原差示光谱法测定总酶含量,通过采用分光光度计法检测GSH浓度的高低来反应GT活力的大小.结果 黄芪总甙+DMH组比DMH组诱导大鼠产生ACF和大型ACF数量均显著下降(P<0.05).DMH组与空白对照组、黄芪组、黄芪+DMH组比较,CYP450酶含量显著降低、GST酶活力显著上升(P<0.05).结论 黄芪总苷有可能通过降低肝微粒体CYP450酶含量、提高GST活力,从而减少二甲肼诱导大鼠结直肠产生异性隐窝灶数量.
