血管生成素和血小板反应蛋白在复发性流产(RSA)患者绒毛组织中的表达及其与血管生成的关系

摘    要:
目的 检测血管生成素(angiopoietin,Ang)、血小板反应蛋白(thrombospondin,TSP)在复发性流产(recurrent spontaneous abortion,RSA)患者绒毛组织中的表达及与血管生成的关系。方法 选取2015年6月至2016年6月于复旦大学附属妇产科医院行流产清宫术的50例患者为研究对象,对照组为正常妊娠行人工流产者,RSA组为≥ 2次自然流产者,每组25例。采用免疫组化SP法检测两组患者新鲜绒毛中Ang-1、Ang-2和TSP-1、TSP-2的表达,并检测CD34阳性和微血管密度(microvessel density,MVD),分析其与血管生成的关系。结果 RSA组绒毛中Ang-1、Ang-2及TSP-2的表达显著高于对照组,TSP-1表达显著低于对照组(P<0.05)。RSA组绒毛组织的MVD值显著低于对照组(P<0.05)。RSA组绒毛组织中Ang-1、TSP-1、TSP-2的表达与MVD呈负相关性,Ang-2的表达与MVD呈正相关性(P<0.05)。结论 RSA患者绒毛组织中Ang-1和TSP-2的异常表达可能与血管生成障碍有关,从而引发RSA。

Abstract:
Objective To detect the angiopoietin (Ang) and thrombospondin (TSP) expression of villus tissues in recurrent spontaneous abortion (RSA) patients and its relationship with angiogenesis. Methods Fifty patients with abortion surgery were selected as the research objects, from June 2015 to June 2016,in Obstetrics and Gynecology Hospital of Fudan University.The control group was normal pregnancy induced abortion,RSA group was more than 2 times spontaneous abortion,25 cases in each group.The Ang-1,Ang-2,TSP-1 and TSP-2 of villius tissues in two groups were detected,and the angiogenesis was analyzed by CD34 positive and microvascular density (MVD). Results The expression level of Ang-1,Ang-2 and TSP-2 in villus tissues of RSA group was significantly higher than that in control group (P<0.05),as the expression of TSP-2 was significantly lower than in control group (P<0.05).The MVD value in villus tissues of RSA group was significantly lower than that in control group (P<0.05).In villus tissue of RSA group,the expression of Ang-1,TSP-1,TSP-2 and MVD showed a negative correlation,while Ang-2 and MVD showed a positive correlation. Conclusion The abnormal expression of Ang-1 and TSP-2 in villus tissue in RSA may lead to angiogenesis limitation,which will cause recurrent miscarriage.
