静脉输液微粒体内分布的模拟研究

摘    要:目的:考察输液微粒进入体内后的主要分布。方法:用微粒计数仪分别检测活性炭和硫酸钡混悬液中的粒径分布,并用其模拟静脉输液微粒分别通过尾静脉给小鼠注射3.62×107,1.13×108,2.72×108个微粒,采用CT断层扫描分析硫酸钡微粒在小鼠体内的分布,以组织器官HE染色观察各组织器官中活性炭的分布情况。结果:活性炭和硫酸钡微粒粒径主要分布在2~10 μm之间。影像学结果显示静脉输入的硫酸钡微粒主要分布在肺脏和肝脏;组织学观察表明活性炭微粒主要分布在肺脏、肝脏和脾脏,少量分布在肾脏和心脏。粒径0~2 μm、>2~4 μm和>4~6 μm的活性炭微粒分别主要分布在脾脏、肝脏和肺脏,肾脏和心脏组织中分别分布着少量2~6 μm和4~8 μm的微粒。结论:粒径<10 μm的输液微粒主要分布在肺脏、肝脏和脾脏,少量分布在肾脏和心脏;在肺脏、肾脏和心脏可观察到极少量粒径≥10 μm的微粒分布。

Abstract:OBJECTIVE To evaluate the primary distribution of infusion particles in vivo.METHODS The particle size distribution of activated carbon and barium sulfate was detected by particle counter, and 3.62×107, 1.13×108 and 2.72×108 particles were injected into mice through tail vein. The distribution of barium sulfate particles in mice was analyzed by CT tomography. The distribution of activated carbon in tissues and organs was observed by HE staining.RESULTS Particle size of activated carbon and barium sulfate were mainly distributed in the range of 2-10 μm. The result of CT indicated that barium sulfate particles were primarily distributed in the lungs and liver. Histological findings showed that activated carbon particles were mainly distributed in the lungs, liver and spleen, and a small amount was distributed in the kidneys and heart. The activated carbon particle size of 0-2 μm, >2-4 μm and >4-6 μm were mainly distributed in the spleen, liver and lungs, respectively, and a small number of particles with the diameter size of 2-6 μm and 4-8 μm were distributed in the kidneys and heart, respectively.CONCLUSION Infusion particles <10 μm were mainly distributed in the lung, liver and spleen, with a small amount distributed in the kidney and heart; very small amounts of particles ≥ 10 μm were observed in the lung, kidney and heart.
