人A组轮状病毒卵黄抗体微球的制备

摘    要:目的:制备抗人A组轮状病毒的卵黄抗体微球。方法:通过单因素和正交试验对人A组轮状病毒卵黄抗体微球的制备工艺进行优选。在模拟胃肠液的环境中,对微球的活性保留率和缓释能力进行评估。结果:最佳制备工艺为:转速为400 r·min-1,海藻酸钠浓度为1.5%,交联固化时间为60 min,初始抗体量为15%,氯化钙浓度为10%,司班80为水相质量的5%,液体石蜡与海藻酸钠水溶液质量比为4:1。微球在pH为2的条件下,2 h后活性保留率达88%以上,6 h累计释放量为50%。结论:成功制备出人A组轮状病毒卵黄抗体微球。

Abstract:OBJECTIVE To prepare yolk antibody microspheres against human group A rotavirus.METHODS The preparation process of group A rotavirus yolk antibody microspheres was optimized by single factor and orthogonal tests. The viability retention and sustained release capacity of the beads were assessed in an environment simulating gastrointestinal fluid.RESULTS The optimal prepatation conditions were as follows:the stirring speed was 400 rmin-1, the concentration of sodium alginate was 1.5%, the cross-linking time was 60 min, the initial concentration of IgY was 15%, the concentration of CaCl2 was 10%, the concentration of emulsifier was 5% and the oil water mass ratio was 4:1. Under the condition of pH 2, the microspheres have an activity retention rate of over 88% after 2 h.The in vitro release study showed that the cumulative release percentage was 50% after 6 h in simulated intestinal fluid.CONCLUSION The microspheres loaded IgY against huaman group A rotavirus were successfully prepared.
