摘    要:
目的:探讨小陷胸汤治疗血脂异常的作用机制。方法:通过在TCMSP平台对小陷胸汤进行活性成分筛选并收集靶点信息;在Genecard,malacard,omim,digsee以及DisGenet等数据库中进行检索,获取疾病靶点;构建韦恩图,得到小陷胸汤治疗血脂异常的潜在作用靶点;并进行GO功能注释和KEGG通路富集分析;构建小陷胸汤中药-成分-靶点-信号通路网络,进行网络特征分析;结合前期研究结果,构建相关网络并分析。结果:共挖掘到小陷胸汤中38种成分及101个潜在靶点,血脂异常疾病1201个相关靶点,得到小陷胸汤-疾病共同靶点50个。共同靶点主要富集于343个生物过程和49条信号通路上。槲皮素可调节PPARA、eNOS,而与PPARA、eNOS相关的30个靶点中有7个靶点参与动脉粥样硬化通路的调控。结论:小陷胸汤可能是通过RELA,IL6,PRKCA,NCOA2,CASP3等靶点,调控动脉粥样硬化通路、AGE-RAGE、TNF-α等信号通路,来调节血脂并预防或延缓AS的发生、发展。

Abstract:
OBJECTIVE To investigate the mechanism of Xiaoxianxiong Decoction in the treatment of dyslipidemia.METHODS Through the TCMSP platform, the active ingredients of Xiaoxianxiong Decoction were screened and the target information was collected; the databases of Genecard, malacard, omim, digsee and DisGenet were searched to obtain the disease targets; the Wayne diagram was constructed to obtain the potential targets of Xiaoxianxiong Decoction in the treatment of dyslipidemia; GO functional annotation and KEGG pathway enrichment analysis were performed; the traditional Chinese medicine-component-target-signaling pathway network of Xiaoxianxiong Decoction was constructed for network feature analysis; combined with the previous research results, the relevant networks were constructed and analyzed.RESULTS A total of 38 components and 101 potential targets were excavated in Xiaoxianxiong Decoction, 1 201 related targets of dyslipidemia diseases, and 50 common targets of Xiaoxianxiong Decoction-disease were obtained.Common targets were mainly enriched on 343 biological processes and 49 signaling pathways.Mistletoe bombesin can regulate PPARA and eNOS, while 7 of 30 targets related to PPARA and eNOS are involved in the regulation of atherosclerotic pathways.CONCLUSION Xiaoxianxiong Decoction may regulate blood lipid and prevent or delay the occurrence and development of AS by regulating atherosclerotic pathway, AGE-RAGE, TNF-α and other signaling pathways through RELA, IL6, PRKCA, NCOA2, CASP3 and other targets.
